“IVF Lab QC: Morphological Sperm Selection and DFI Management․
Selecting high-quality sperm is crucial for successful fertilization and blastocyst development in ICSI cycles. Standard sperm preparation methods often fall short, particularly in cases with a high Sperm DNA Fragmentation Index (DFI).
Sperm Selection Methods: A QC Comparison
- Density Gradient Centrifugation (DGC): Isolates highly motile sperm, but high-speed centrifugation carries a risk of generating Reactive Oxygen Species (ROS).
- Swim-Up Technique: Delivers a clean yield for low-DFI samples, but recovery rates drop significantly in cases of severe oligo-asthenozoospermia.
- Microfluidic Sperm Sorting (Chip Method): A single-step, non-centrifugation process that minimizes double-stranded DNA damage, leading to superior blastocyst conversion rates in high-DFI cases.
- IMSI / Physiological Selection (PICS): Utilizes hyaluronic acid binding or 6000× magnification to select mature, vacuole-free sperm.
Key QC Considerations for Embryologists
- Centrifugation Speed and Time: Avoid excessive force (>300g) to prevent sperm membrane damage and oxidative stress.
- Holding Temperature: Maintain prepared sperm samples at optimal temperatures to prevent a decline in motility.
