Arshhita A.: 3PN Oocytes – Why Does Fertilization Go Wrong?

Arshhita A., Embryologist at Generation Fertility, shared on LinkedIn:

”3PN Oocytes: Why Does Fertilization Go Wrong?

In IVF, normal fertilization typically shows 2 pronuclei (2PN) – one maternal and one paternal.

But what happens when we observe 3 pronuclei (3PN)?

Possible Causes

  • Dispermy – two sperm fertilize one oocyte
  • Failure of second polar body extrusion
  • Abnormal oocyte activation or chromosome segregation
  • Timing-related assessment difficulties

Why Does It Matter?

A 3PN zygote may have an abnormal chromosome complement, including triploidy. Although some 3PN embryos can develop into blastocysts, blastocyst formation does not guarantee genetic normality.

An Important Lesson for Every Embryologist

Beautiful morphology doesn’t always mean normal genetics. Every pronucleus tells a story about fertilization – and accurate assessment matters.

In embryology, development is important, but chromosome integrity matters too.”
