Karthik Raj: True Biological Embryo Mosaicism vs. Technical Detection

“Embryo Mosaicism: True Biological Mosaicism vs. Technical Detection
A mosaic PGT-A result does not necessarily provide a complete picture of the embryo’s chromosomal constitution.
Biological mosaicism arises when an embryo contains cell populations with different chromosomal complements, often due to post-zygotic mitotic errors.
But the reported result also depends on the technology used to analyse a small trophectoderm biopsy, which may not fully represent the inner cell mass or the entire embryo.
Amplification bias, analytical noise and thresholds used by laboratories to classify intermediate copy-number signals can contribute to a mosaic result.
This creates an important distinction:
A reported mosaic result is not always equivalent to confirmed whole-embryo mosaicism.
Understanding this difference is essential when interpreting PGT-A findings, counselling patients, and considering embryo-transfer decisions.
The challenge is not simply detecting a signal – but understanding what that signal truly represents biologically.”
