Sean Lauber: NIPT and Amniocentesis after PGT-A Reveal Few Abnormalities

Sean Lauber, Founder of Remembryo, shared on LinkedIn:

“A new study found that amniocentesis detected a chromosome abnormality missed by PGT-A in just 0.1% of ongoing pregnancies after euploid embryo transfer.

PGT-A is a screening test, not a diagnostic test. Even when an embryo is identified as euploid, it doesn’t guarantee that the developing baby will have a normal number of chromosomes. PGT-A only tests a few cells that may not represent the entire embryo.

For these reasons, prenatal testing is generally recommended after PGT-A, including NIPT screening or amniocentesis.

But how often do these tests find abnormalities after euploid transfer?

Lu et al. (2026) examined prenatal testing results from 1,079 ongoing pregnancies after euploid embryo transfer (over 12 weeks pregnant).

Among 762 pregnancies tested by amniocentesis, only one (0.1%) had an abnormality PGT-A was expected to detect but missed. Three other abnormalities were found that were at or beyond the detection limits of the PGT-A method used.

There were eight high-risk NIPT results, but seven were found to be normal on amniocentesis, showing that most high-risk NIPT results were false positives in this study.

Overall, chromosome abnormalities were rare after euploid transfer based on NIPT and amniocentesis.
