Efficient sperm preparation is crucial for isolating motile, morphologically normal, and DNA-intact spermatozoa — essential for fertilization success in both IVF and ICSI.
The choice of technique depends on semen parameters and the clinical scenario.
Major Techniques:
- Swim-Up Method/direct layering
• Relies on sperm motility for separation.
• Best suited for normozoospermic samples. - Density Gradient Centrifugation (DGC)
• Separates sperm based on density and morphology.
• Effective for samples with debris or low motility. - Simple Wash Method
• Removes seminal plasma quickly.
• Common in donor sample processing. - Microfluidic Sperm Sorting
• Mimics natural sperm selection in the female tract.
• Reduces oxidative stress and improves DNA integrity.
Quality Control Considerations:
Maintain sperm viability and motility throughout the procedure.
Control centrifugation force, temperature, and pH.
Routinely check media osmolality and sterility.
