The choice of technique depends on semen parameters and the clinical scenario.

Major Techniques:

  1. Swim-Up Method/direct layering
    • Relies on sperm motility for separation.
    • Best suited for normozoospermic samples.
  2. Density Gradient Centrifugation (DGC)
    • Separates sperm based on density and morphology.
    • Effective for samples with debris or low motility.
  3. Simple Wash Method
    • Removes seminal plasma quickly.
    • Common in donor sample processing.
  4. Microfluidic Sperm Sorting
    • Mimics natural sperm selection in the female tract.
    • Reduces oxidative stress and improves DNA integrity.
