“Sperm DNA Fragmentation (SDF): What It Means for Male Fertility
While most fertility checks focus on sperm count, motility, and morphology – there’s another important factor that often goes unseen: DNA quality.
Sperm DNA Fragmentation (SDF) measures the integrity of the sperm’s genetic material. High SDF levels can reduce fertilization, embryo development, and pregnancy success – even when standard semen results look normal.
A 2025 review of 174 studies by Joaquín Llácer et al. found that:
- High SDF is associated with lower chances of natural conception and IUI success.
- It’s linked to poorer fertilization and embryo quality, and higher miscarriage risk.
- It can often be improved through lifestyle changes, antioxidants, or varicocele repair.
However, results vary – there’s still no universal cutoff or standardized test, and findings can depend on egg quality and female age.
That’s why experts recommend SDF testing as a second-line tool: valuable for couples with unexplained infertility, recurrent miscarriage, or repeated IVF failure.
