Kabita KB: What Happens if PLCZ1 Is Defective?

Kabita KB, Senior Embryologist at IHR Guwahati, Assam, shared on LinkedIn:

“PLCZ1 Gene:

The Molecular Key to Sperm-Induced Oocyte Activation and Successful Fertilization After ICSI.

PLCζ1 is not itself the activation signal; it encodes the sperm protein phospholipase C zeta (PLCζ), which initiates the intracellular signalling cascade that activates the oocytes.

What happens if PLCZ1 is defective?

If the sperm has insufficient PLCζ protein, or the protein is dysfunctional, the calcium oscillations may fail to occur or may be inadequate.

  • Possible clinical outcome: total fertilization failure (TFF) even after ICSI.
  • Underlying mechanism: the sperm may be injected successfully, but the oocyte does not receive the necessary activation signal.
  • Important distinction: a PLCZ1 defect is one possible cause, not the only cause of ICSI fertilization failure.

Oocyte-related activation defects and other sperm factors can also contribute.

Core messages: PLCZ1 encodes the sperm protein that triggers the IP₃–Ca²⁺ signalling pathway. Without an adequate activation signal, an ICSI-injected oocyte may fail to complete activation and fertilization.”
